In the final step of' mitochondrial division,
outer and inner division apparatuses are loosely associated in a red alga
Cyanidioschyzon merolae.
Mitochondrial matrix (red), FtsZ ring (green), and Dynamin ring (blue) were respectively
immunolabeled with the antisera against EF-Tu(mt), CmFtsZl or CmDnml,and visualized with secondary antibodies conjugated with Alexa555, Alexa488
or lexa350. Images were observed under a fluorescence microscopy and digitally
captured and reproduced through a cooled 3CCD camera system. The phase contrast
image was faintly overlaid on the merged image. Bar: 1µm
While mitochondrial FtsZ is thought to maintain the placement of mitochondrial division
site at inside of the inner membrane, dynamin is recruited for the final severance
of mitochondria to the mitochondrion dividing ring (MD ring) at the outside of
the constricted outer membrane. The fact that the localizations of the FtsZ
(green) and the dynamin (blue) do not tightly associated with each other suggest
that the MD ring but not FtsZ mediates the placement of the final severance
diven by dynamin (see Nishida K., Misumi O., Yagisawa F., Kuroiwa H., Nagata T.
and Kuroiwa T. 2004: J Histochem. Cytochem., 52(7): 843-849).
