Northern blot analysis of BLTR mRNA in HL-60 and U-937 cells
The BLTR mRNAs are induced in the presence of 1 micro M RA in HL-60 cells.
Poly-A+ RNA (3 micro g) from HL-60 and U-937 cells were electrophoresed
in a 1% agarose gel, and transferred to a Hybond-N nylon membrane (Amersham).
The filter was hybridized with [32P]-dCTP-labeled ORF of the HL-5 clone
or a human glutaraldehyde 3-phosphate dehydrogenase cDNA (Clonetech) in
hybridization buffer containing 4 x SSC, 5 x Denhardt's solution, 0.2% SDS,
200 micro g/ml salmon sperm DNA, 50% formamide at 42 ¡C for 24 h. The filters
were washed in 0.1 x SSC, 0.1% SDS at 65 degree and subjected to autoradiography.
